Is it possible to make a dervish assassin as your starting 2nd pro? If not when will I be able to later in the game. (Own all the GuildWars)

No, you cannot choose Assassin as your starting secondary in Nightfall. You will be able to switch secondaries to any profession, including Assassin, after reaching the Sunspear Sanctuary and completing [wiki]Building the Base: Prisoners of War[/wiki].
